In this assay, mitotic cells are selectively identified using an antibody specific for a phosphorylated core histone protein that is abundant in the nuclei of dividing cells. Many therapeutic agents target cancerous cells by blocking mitotic progression, eventually causing cell death. Compounds that inhibit mitotic progression, such as nocodazole, vinblastine, colchicine, paclitaxel and curacin, increase the mitotic index of a cell population. The Mitotic Index Kit assay can be used both for primary screening of compounds with anti-mitotic activity and secondary screening for functional analysis of lead compounds. The Mitotic Index Kit, in combination with the ArrayScan HCS Reader and the Mitotic Index BioApplication software enables automated plate handling, focusing, cell image acquisition, analysis and quantification. Mitotic index, a valuable means of characterizing cell proliferation, represents the cell fraction within a population undergoing cell division. The mitotic index is often higher in cancerous cells because of uncontrolled cell proliferation. Mitotic cells are visually identifiable by the presence of mitosis-specific proteins. Standard assays determine mitotic index either manually through direct microscopic characterization of tissue samples, or indirectly through analysis of DNA content using fluorescence-activated cell sorting. The Mitotic Index Reagent Kit used in conjunction with the Mitotic Index BioApplication Software (Product No S50-0005-1) and the ArrayScan HCS Reader, enables objective and automatic mitotic index quantitation on a screening scale much faster than manual microscopy methods.
